Synthesis and characterization of solid lipid nanoparticles loaded with Fomes fomentarius polysaccharides as a potential in diabetes treatment abstract

Common diabetes medications have many side effects. Ethanol extract from a culture medium of Fomes fomentarius has shown anti-inflammatory and hypoglycemic effects due to polysaccharides such as betaglucans that increase PI3K/Akt activity. However, low solubility and chemical or enzymatic degradation lead to reduce polysaccharides efficacy. Controlled delivery of these polysaccharides improves its therapeutic effect and bioavailability. Among delivery systems, solid lipid nanoparticles (SLNs) have shown many advantages, such as biocompatibility and ease of manufacturing processes. In this study, solid lipid nanoparticles containing polysaccharides were prepared by the microemulsion method. The mean particle size and the zeta potential of SLNs were 203.1 nm and -25.5 mV, respectively. The drug entrapment efficiency (EE) and loadingefficiency (LE) were determined using UV-visible spectroscopy were reached up to 93.6 % and 9.27%, respectively. In vitro release studies showed a sustained release of polysaccharides from SLNs. FTIR study of polysaccharide loaded SLN showed the main peaks of polysaccharide and lipid, indicating no chemical interaction between drug and lipid. TGA analysis showed appropriate thermal stability of SLNs and good incorporation of polysaccharides in SLNs.
