Fabrication and Optimization BSA Nanoparticles as Drug Delivery Systems abstract
Nanoparticle prepared by coacervation method and subsequent cross-linking of nanoparticle represent promising carrier for drug delivery. Bovine Serum Albumin (BSA) nanoparticles with a size from 78.9 to 421 nm diameter were prepared using a self-assembly simple coacervation method and cross-linking with glutaraldehyde. Nanoparticle was purified with 48,800 g centrifuge following by dialysis, micro and ultra filtration. The produced nanoparticles were subjected to physical, morphological as well as biochemical characterisation. Photographs obtained for image analysis studies by SEM and AFM proved semi-spherically shape of the particles whilst size distribution was measured by photon correlation
spectroscopy. Fourier transforms infra-red spectroscopy (FTIR) and gel electrophoresis results indicated the nanoparticles have high purity. Several process parameters were examined to achieve a suitable size of nanoparticle through one factor at a time method such as pH, temperature, BSA concentration, agitation speed, glutaraldehyde concentration, organic solvent adding rate and the ratio of BSA/ organic solvent. The minimum size of nanoparticles at de desired incubator of 4°C, pH=7.5, 30 mg.ml-1 BSA concentration and constant agitation speed of 500 rpm was obtained. The mechanistic fabrication of protein nanoparticles and their characterisation is strongly discussed.
