Real Time PCR to Detect Contamination of Animal and Poultry Feedwith Salmonella in Tehran

Abstract:

Salmonellosis is one of the most serious health problems of human societiesand Salmonella, the causal agent of this zoon tic disease, is transmitted via thefecal-oral route. Finding the transmission route of Salmonella makes itpossible to control or stop its pathogen city. For this purpose, the presentstudy was conducted to use two methods (the conventional culturing methodand Real Time PCR) for determining the extent of Salmonella infection inanimal and poultry feed in Tehran Province. DNA was first extracted usingthe enzymatic method, and Salmonella bacteria were detected in seven feedsamples by employing the invA gene and Real Time PCR. Based on thestandard curve and on the obtained results, Real Time PCR, with thesensitivity of 100%, could identify bacterial concentration of 10 CFU/ml inthe samples, and, compared to the culturing method, its specificity was98.37%. Considering the time-consuming nature of the conventional culturingmethod and the problems associated with the conventional culturing, RealTime PCR and the invA gene, as the specific indicator of Salmonella, wereemployed as a substitute for the conventional culturing method in thisresearch. Moreover, the specific primer Salmonella Centrica was developedand patented by the author of this article in 2014. This research was carried out from late September 2012 to late April 2013. Sixty-seven samples ofanimal and poultry feeds, 34 samples of concentrates, from silos, of alfalfa,and from feed mangers at dairy farms, and 33 samples of pelleted and groundfeeds from poultry farms and factories producing animal and poultry feeds inTehran Province were used in this study.
