Abstract:

HPA-1 is one of antigen on platelet membrane with two codominant form a & b. HPA-1disparity between blood donors and recipients makes blood transfusion reactions. There aredifferent methods to detect HPAs on platelet with different sensitivity. So the aim of thisstudy was to compare ELISA & Molecular method (PCR) to evaluate HPA-1 on plateletsderived from blood donorsDNA was extracted from 3 ml of whole blood in EDTA collected from 100 Iranian blooddonors. Human Platelet Alloantigen-1 (HPA)-1 genotyping were performed by thepolymerase chain reaction –sequence-specific primer techniques (PCR-SSP) and HPA-1aphenotyping was performed by ELISA methodThe frequency of HPA-1 genes were: HPA-1a 0.98, HPA-1b 0.02. HPA-1a phenotyping byELISA showed: positive in 65%, negative in 10% and indeterminate in 25% of the samples.Obtained results weren t concordant in total. ELISA method may offer efficient HPA-1typing in large numbers samples but may be reliable and have accurate results just in HPA1anegative samples.
