جداسازی و تخلیص آنزیم گلوتاتیون- اس- ترانسفراز ‭GST ‬از انگل بالغ فاسیولا

‭Fascioliasis due to infection with the trematode parasite fasciola is a disease of 1‬ص ‭ economic and health importance in sheep I cattle and humman the potent,ial of GST as a vaccine candidate against liver fluke infection in ruminants was studied , by others. Adult parasites were obtaind fresh from the bile clucts of infected sheep at local abattoirs. After washing in PBS , the adult worms ( Fasciola gigantica) were stored at -70 oc. Adult worms were homogenised and suspension was centrifuged . supernatant was used for the purification of GST. GST was purified from adult worm extract by affinity chromatography on GSH- agarose beads. eluted material was electrophoresed. we found that the GST fraction comprised two major components of 25- 28 KDa. Enzyme activity of GST was assayed by measuring enzymic conjugation of glutation with 1. chiaro -2,4 dinitrobenzeue.‬
