Abstract:

Application of new plant propagation methods such as tissue culture can remove Citrumelo propagation obstacles (e.g. fewer seeds production). In this method, Citrumelo was propagated in vitro from adventitious bud (1cm) and leaf (cm2) explants (taken from cutting plants). Explants were cultured on: 1/2MS (T1-1), MS (T1-2), MS supplemented 0/5 mg/l of NAA and 0/5 mg/l of BA (T1-3), MS supplemented 0/5 mg/l of NAA and 1 mg/l of BA (T1-4), MS supplemented 1 mg/l of NAA and 0/5 mg/l of BA (T1-5 ), MS supplemented 1 mg/l of NAA and 1 mg/l of BA (T1-6), 1/2MT (T2-1), MT (T2-2), MT supplemented 0/5 mg/l of NAA and 0/5 mg/l of BA (T2-3), MT supplemented 0/5 mg/l of NAA and 1 mg/l of BA (T2-4), MT supplemented 1 mg/l of NAA and 0/5 mg/l of BA (T2-5), MT supplemented 1 mg/l of NAA and 1 mg/l of BA (T2-6). The maximum percentage of callus formation, adventitious shoots and roots growth were obtained by axillary's bud (with comparative of leaf explants) in MS medium (comparative with MT). The best results for the callus production were achieved in the T1-5 medium while the best results for the shoot production were achieved in the T1-6 medium. By subculturing new shoots production in T1-2 (for the shoot growth) and T1-1 (for the root production and growth), a multiplication rate 6:1 was attained every two month. In vitro produced plantlets were transplanted to autoclaved perlite and watered with MS mineral solution. Transplanted plantlets were covered with polythene sheets to maintain high relative humidity.
