Evaluation of Allogenicity effects of Human Amniotic Epithelial Cells on Peripheral Blood Mononuclear Cells from Healthy Subjects

Abstract:
Background: Human amniotic epithelial cells (hAECs) have pluripotency, repair capacity, immunomodulatory properties, differentiate into multiple cell lineages. Therefore, we investigated allogenicity effects of hAECs, as a potential therapy for cell therapy, on peripheral blood mononuclear cells (PBMCs) from Healthy Individuals.Methods: PBMCs isolated from 4 healthy donors using density gradient centrifugation. hAECs were separated from 4 amnions delivered by healthy women. Isolated-hAECs from 4 amnions were mixed with each other. PBMCs (2.5 × 104) were co-cultured with hAECs at different ratios (1:1, 1:2, 1:3) along with positive (1:0) and negative control(1:0), for 3 days. In positive control, the Proliferation of PBMCs was stimulated with anti CD3/CD28 (1μg/ml). BrdU cellular ELISA kit used to proliferation assay.Result: In the positive control, the proliferation of PBMCs were significantly more than 1:1, 1:2 , 1:3 ratios(PConclusion: These findings suggest hAECs are not the allogenicity properties and may exert inhibitory activities on PBMCs in vitro. Therefore, the possibility of rejection of these cells can decrease after transplantation as a potential cell source for generative medicine.
