Application of chemiluminescence and ELISA methods for toxoplasmosis Seroepidemiological study in hemodialysis patients abstract
Introduction: Toxoplasma gondii is the most common opportunistic infection among people with impaired immune systems, such as
hemodialysis patients which mainly occurs in tropical and semi-tropical with high humidity. This study aimed to investigate IgM and IgG in
hemodialysis patients applying ELISA and
chemiluminescence methods.
Toxoplama infection in
hemodialysis patients is high and they are in high risk of severe damage, therefore, results of this research, in planning for control and prevention of parasite can be of help. Materials and Methods: A cross-sectional study was carried out in studied population of 80
hemodialysis patients as subjects and 80 healthy individuals as controls by obtaining written consent from both groups. 5 ml blood was taken, centrifuged and 2 ml of serum isolated, placed in micro-tubes and kept in _20°C until testing. The diagnostic ABON (UK) kit was used to measure Toxoplasma IgG and IgM evaluating through Ab Capture ELISA and Indirect ELISA methods. Chi-square tests and SPPSsoftware version 21 were used for statistical analysis. Results: In ELISA, a total of 57 positive IgG was detected in two groups of
hemodialysis patients and healthy individuals however, this was positive in 38 subjects (both groups) by chemiluminescence. In fact, 19 positive cases in ELISA, were negative by chemiluminescence. Positive IgM was detected in 9 cases by ELISA while this was 8 cases in
chemiluminescence method. In fact, one case was positive in ELISA but negative in chemiluminescence; Table 1. Conclusion: The results showed that the prevalence of Toxoplasma in
hemodialysis is higher than healthy subjects. It is therefore recommended to relevant authorities in place plans in order to control and prevent the parasite in
hemodialysis patients. Chemiluminescence compared to ELISA is accurate and sensitive with very low false positives. In fact, IgM and IgG in patients and healthy individuals for a total review of 20 cases were positive in ELISA, where got one negative in
chemiluminescence method.Table 1. IGg detection by Chemiluminescence and ELISA in
