Cytotoxicity of TPSF Small Molecule on MCF-7 Breast Cancer Cells in Hydrogel Scaffold

Abstract:
Introduction and Aim: Breast cancer is claimed to be common cancer amongst women. Main procedure of treatment is chemotherapy that has side effects. The use of small molecules has increased in cancer treatment nowadays. One of these small molecules is TPSF. Also, the enormous versatility of hydrogel materials makes it possible to design scaffolds with suitable mechanical properties to biomimetic modeling. In this study, we investigated the apoptotic and cytotoxicity effects of TPSF on the MCF-7(Michigan Cancer Foundation-7) in the 3D fibrin hydrogel scaffold. Methods: we seeded MCF-7 cells at a density of 50×103 into 24 well plates in the fibrin hydrogel and then we exposed them to concentrations of 0/5, 1, 2, 5, and 10 μM of TPSF. Fibrin hydrogel was prepared by polymerization of fibrinogen (3mg/ml) by thrombin (120U/ml). MTT assay was used to evaluate cell viability. Also, Giemsa and Acridine Orange/Ethidium Bromide (AO/EB) staining were used for apoptosis detection. Results: We determined IC50 concentration for TPSF (5 μM,) which induced 50% cell death and our observations showed a significant decrease in cell viability of treated cells with IC50 concentration of TPSF to the extent of 50, 38 and 27% after 1, 3, and 5 days respectively, in a dose and time-dependent manner. Also, apoptotic morphology in treated cells confirmed by AO/E and Giemsa staining. Conclusion: Therefore, we suggest that TPSF exerts significant cytotoxic effects and can be used as a new therapeutic target for breast cancer treatment.
