تعیین تیتراژ واکسن ‭MMR ‬به روش میکرو و مقایسه آن با کشت در لوله آزمایش

Virus titration in products including measles, mumps or rubella is very important, because of availability of effective immunity following vaccination with monovalont or MMR vaccine. Based on the last manual of WHO about potency test, the micro method has been recommended instead of the tube method. In this study, at first the proper samples of monovalent vaccines of measles, mumps, rubella and also MMR were selected as in-house standards. Then they were titered in tube. The standard vaccine, in addition to test sample of each virus, were used in micro method. Study on different details of micro method were done and then the method for measles potency was set up in a microplate ; for each well:0.1cc of 2 105 vero cell/ml, 50 of viral dilution, 50 of maintenance medium. The method was certified for mumps virus, both in-process and final products, of course with using complementary test of hemadsorption. Finally the same procedure with RK-13 cell concentration of 1.6 105 /ml was accepted for rubella virus, too. Difference of titer between plate and tube was <0.5 log. In next process, each of mumps, measles and rubella antisera were titered in plate. Then two antisera with titer 1/400 x 1/200 were used for seroneutralization the two viruses and titration of third virus, in MMR. Also identification test for MMR was done with the process.
