Effect of Thioflavin–T on obesity in NMRI male mice

Abstract:

Obesity has become a worldwide health problem. Most of the synthetic anti-obesity drugs have failed to the management of obesity due to either ineffectiveness oradverse effect. The main of the present study was intended to examine the effects of thioflavin T on a high-fat diet (HFD) induced obesity on body weight and adipocytes size in NMRI mice model. Male NMRI mice were fed HFD for 8 weeks to induce obesity while one group served as the control that received normal rodent diet. After 8 weeks of feeding with high-calorie ingredients, the mice exhibited significantly (p<0.001) enhanced body weight gain. Thioflavin T dissolved in water (12 mg/kg) administered orally one day to HFD fed mice for 8 weeks and an obese group didn’t receive any treatment. Alteration in body weight gain was assessed. At the end of treatment with thioflavin-T was measured body weight, in obese mice compared with mice fed with normal diets and fat tissue in order to prepare microscopic slides was fixed in formalin. Thioflavin-T treatment showed significantly (p< 0.001) decreased body weight gain. This compound leads to a reduction in the size of adipose tissue cells to some extent. In conclusion, the tested compound was able to prevent HFD induced obesity in mice model and attenuation of visceral fat accumulation.
